Nuciferine ameliorates high‐fat diet‐induced disorders of glucose and lipid metabolism in obese mice based on the gut–liver axis

High-fat diets (HFDs) can trigger lipid accumulation, insulin resistance, and inflammatory responses in the body, causing glucose metabolism dysfunction. This study aimed to explore role of nuciferine on HFD-induced disorders obesity mice models, further identify possible underlying mechanisms. In our experiments, effectively ameliorated resistance by IRS/PI3K/AKT pathway, alleviated accumulation via AMPK-mediated regulation FAS ATGL expression. Nuciferine also suppressed intestinal pathological damage, as evidenced improvement barrier integrity increasing expressions tight junction proteins claudin, ZO-1, occludin. addition, 16S rRNA gene sequencing fecal microbiota suggested that changed diversity composition gut HFD-fed mice. decreased ratio Firmicutes/Bacteroidetes, relative abundance LPS-producing genus Desulfovibrio bacteria which involved metabolism. summary, obese balancing gut–liver axis.
